Staining polystyrene microspheres (Diameter: 6 um) with two different fluorescent dyes, each at 10 concentrations, results in up to 100 differently fluorescence encode microspheres.
Binding antibodies of different substances to be tested to specific encode microspheres through covalent crossing ensures that each fluorescent microsphere corresponds to a detection target.
Mix the fluorescence encode microspheres of different detection targets. Each microsphere will serve as an independent carrier of bio-reaction. Then add the analyte, biotinylated antibodies, and
fluorescein-labeled allophycocyanin sequentially in stages to form a 'sandwich' structure.
The dyed microspheres are sequentially carried through the fluidic system by the sheath fluid. The red light source (635nm classification laser) in the instrument measures the dye in the microspheres and identifies the microsphere code and analyte type, and the green light source (532nm reporting laser) measures the fluorescence intensity of the microsphere reporter molecules. After analysis, the fluorescence signal is converted into a quantitative concentration detection result.
